This will show you how to create the 3D "Jello Text". The plugins needed to do this are the Feather Plugin and the Transparency Plugin, which can be found in the Paint.NET Official Forum. The end result will look like this:

To start off, start with a new image, any size.
Create a new layer. Turn off anti-aliasing and write in your text in a color of your choice. Name this layer "Text".

Using the magic wand , select all of the transparent areas around the text and invert the selection(Ctrl+I). Create a new layer and choose a lighter version of the color you used for your text as your primary color and a completely transparant color for your secondary color. Using the gradient tool, create a radial gradient starting from under your selection to above it.

Merge this layer into the text layer. Duplicate the text layer and name the lower one 3D. Do a motion blur on the 3D layer at a -90 degree angle, and make sure that "Centered" is off. The distance of the motion blur depends on how thick you want the letters to be. Using the Transparency plugin, adjust the transparency to be 100% opaque.

On the text layer, select all the transparant area around the text. Create a new layer, name it "White", and fill the selection with white. Feather the White layer by 10.

Once again, on the text layer, select the transparant areas. Switch to the White layer and delete the selection. Merge the White and Text layers.
Switch to the Text layer and select all the transparent areas. Invert the selection (Ctrl+I) and create a new layer. Name this new layer "Gloss". Using the Rectangle tool set at Draw Filled Shape , create a white rectangle in the selection. Make it semi-transparent using the Transparency plugin. Merge the Gloss and Text layers together.
Merge the Text and 3D layers together and apply a Glow to finish.
